MATRIXYL 3000

Matrixyl^® is a trade mark of Sederma SAS. It designates an anti-wrinkle active ingredient for cosmetics containing a specific

matrikine which cosmetic nomenclature denomination is palmitoyl pentapeptide-4 (palmitoyl pentapeptide-3 before 2006).

It contains a peptide of 5 aminoacids linked to a 16-carbon chain for improving the penetration of the molecule through the

lipidic structures of the skin.

Matrixyl 3000 combines palmitoyl oligopeptide and palmitoyl tetrapeptide-7 not just because two active ingredients are better

then one. According to the Sederma corporation, these two peptides produce a synergy when used together, i.e.

the skin benefits of such combination are greater than what one would expect if each agent's effects were independent of each other.

Here's a fictitious example of synergy to illustrate my point: Ingredient A causes a 10% improvement and ingredient B causes

a 15% percent improvement whereas A+B combo causes a 60% improvement (as opposed to the expected 25%

improvement).

Sederma also reported a small human study involving two groups of 24 volunteers


where Matrixyl 3000 performed considerably better than placebo and somewhat better than "classic" Matrixyl,


including the following beneficial effects after two month of daily application ^*:

ARGIRELINE

Acetyl hexapeptide-3 is marketed as Argireline by the Barcelona-based research laboratory Lipotec.

Argireline is a peptide which is a fragment of SNAP-25, a substrate of Botulinum toxin (Botox).

It prevents the formation of expression wrinkles by somehow inhibiting muscle movement.

For this reason, it is sometimes called Botox in a jar.

Argireline decreases the visible effects of aging by reducing the deep wrinkles and lines that occur 

around the forehead and eyes.

The first peptide to reduce expression lines, an alternative to topical botox injections, 

it works the same as botulinum toxin but safer and smoother. 

This anti-wrinkle peptide, the first of its generation, diminishes wrinkles and expression lines.

The manufacturers of Argireline carried out a study on a group of female volunteers.

The volunteers used a cream that contained 5% Argireline for 30 days. 

At the end of 15 days of continuous use, lines around their eyes were reduced by 17%. 

After 30 days, lines and wrinkles were diminished by 27%.

Another independent study published in the International Journal of Cosmetic Science

found that Argireline reduced wrinkles by 30% after 30 days of use.

ADENOSINE

Adenosine is endogenously present in all cells of the body. As a drug, it is only given in a hospital setting.

The use of adenosine in cosmetic products has expanded the last ten years due to

the discovery of the anti-wrinkle/anti-aging properties of adenosine.

As we are of the opinion that bio active compounds used in cosmetic products should not cause adverse 

effects nor exert a prominent biological effect like influencing with the functioning of the hart.

Therefore, we chose to use the lowest effective drug dose divided by three as a NOEL value.

We have estimated the margin of safety (MoS) for six different cosmetic product categories that contain adenosine; 

anti-aging face cream, anti-aging eye cream, anti-hair loss product, body lotion, hand cream and tanning oil. 

Since the NOEL is based on human data, a MoS of 10 is sufficient as a safety margin.

More than a decade ago, scientists discovered that adenosine—a compound that regulates heart rhythm—also helps

skin cells function and grow.

Now cosmetics researchers have proven that adenosine is an effective ingredient in wrinkle creams

After two months of twice-daily treatments, both of the adenosine products increased skin smoothness and reduced

deep wrinkles in the eye and forehead areas, while the placebo had no beneficial effect. Surprisingly, 

big improvements occurred on coarse wrinkles after three weeks of treatment. One contributor to expression 

lines may be contraction of skin cells,

HYALURONIC ACID

Hyaluronic Acid (HA) is a polysaccharide and a compound that occurs naturally in every tissue of the body, 

with the highest concentrations occurring in tissues such as skin, cartilage and eyes.

Providing protection to cell structures, HA is the natural gel between skin cells that serves as a water 

reservoir for the cells.

Niacinamide (Vitamin B3)

Niacinamide (also known as nicotinamide) is a compound derived from niacin (vitamin B3).

It has been shown to exert anti-inflammatory action within the skin. In a randomized, controlled clinical

trial of 65 subjects, 5% nicotinamide performed comparably to 2% clindamycin (both as topical

formulations) in reducing acne severity scores.

In a Japanese double-blind, placebo-controlled trial examining sebum production, fifty subjects received

a topical 2% nicotinamide moisturizer daily and fifty subjects received a placebo.

After two weeks of treatment, sebum production of the nicotinamide group was reduced significantly in

comparison to the placebo group.

A separate randomized, double-blind multicenter trial compared a 4% nicotinamide emulsion to a 1%

clindamycin formulation, both applied daily as topical formulations.

The nicotinamide formulation was found to be superior in terms of safety, tolerability, and efficacy.
